Guidelines for Submitting 10x Samples to the Genomics Core
Samples for Library Preparation
Submission for 10x Chromium X
Single Cell 3' Expression Library
Please contact the Genomics Core 2 weeks prior to the initiation of your single cell expression project. Researchers must coordinate their sample delivery with core personnel prior to submission. The cell sample must be submitted as a completely dissassociated cell suspension with a high viability percentage of >80%. Please use the following link to access the best practices for sample preparation and access to supporting documents and training videos for cell preparation and single cell expression project planning.
Single Cell 5' Expression Library and V(D)J Enrichment Library
Please contact the Genomics Core 2 weeks prior to the initiation of your single cell expression project. Researchers must coordinate their sample delivery with core personnel prior to submission. The cell sample must be submitted as a completely dissassociated cell suspension with a high viability percentage of >80%. Please use the following link to access the best practices for sample preparation and access to supporting documents and training videos for cell preparation and single cell expression project planning.
Single Cell ATAC Chromatin Library
The success of a single cell ATAC sequencing project is determined by the quality of the cell nuclei preparation provided to Genomics Core. Researchers must coordinate their sample delivery with core personnel at least 2 weeks prior to submission. The nuclei sample must be submitted as a completely dissassociated nuclei suspension derived from cells with a high viability percentage >80%. Cryopreserved cells are acceptable for nuclei isolation. The 1X Nuclei Buffer required for nuclei resuspension will be provided by the Genomics Core personnel to support your Single Cell ATAC project.
Single Cell Multiome ATAC + Gene Expression
Single cell multiome ATAC + gene expression requires cell nuclei to initiate the multiome library preparation. The success of a single cell multiome ATAC + gene expression sequencing project is determined by the quality of the cell nuclei preparation provided to the Genomics Core. Researchers must coordinate there sample delivery with core personnel 2 weeks prior to submission. The nuclei sample must be submitted as a completely dissassociated nuclei suspension derived from cells with a high viability percentage > 80%. Cryopreserved cells are acceptable for nuclei isolation. The 1X Nuclei Buffer required for nuclei resuspension will be provided by the Genomics Core personnel to support your single cell multiome ATAC + Gene Expression project.
Chromium Gene Expression Flex - Fixed RNA
Chromium Gene Expression Flex - Fixed RNA single cell can be performed using fresh, frozen and FFPE fixed samples. Singulated cells are required to initiate the protocol. Tissue or cells can undergo a fixation protocol allowing for -80C storage prior to the Chromium processing of the singulated cells or nuclei allowing the workflow to be optimized for multiple sample management or time course studies. Samples can be batched with multiplex barcoding. Researchers must coordinate there sample delivery with core personnel 2 weeks prior to submission. The cells or nuclei must be submitted as a completely dissassociated suspension.
