Transgenic Mouse Production

 

Services provided by the facility

  • Use F1 hybrid mouse embryos FVB x C57Bl/6 unless requested by the user and approved by the facility.
  • The facility will superovulate donor mice, harvest single cell embryos and inject DNA provided by the investigator.  Surviving embryos will be implanted into pseudopregnant females.
  • Plasmid DNA will be provided to the facility were it will be linearized and purified.  The DNA will then be used for injection into embryos and implanted. Genotyping will also be provided by the facility to identify transgenic founders.  A guarantee of at least two transgenic founders will be provided for each construct. It should also be noted that we can not guarantee expression and/or transmission of the transgene
  • The facility is now offering the service of BAC DNA preparation. The facility will grow up a bacterial culture, isolate and prep the BAC for microinjection in the appropriate buffer. The facility will also genotype pups for one gene of interest and will guarantee two founders positive for that one gene.

Responsibilities of the investigator

  1. Discuss experimental design with facility director before initiation of the project. Important, when designing your vector, ensure the transgene sequence is flanked by unique restriction enzyme sites. This allows for easy removal of vector backbone (if left on, the vector backbone sequence can interfere with expression of your transgene) when preparing the DNA for injection.
  2. Provide 25ug of your circular DNA construct at a concentration of 1ug/ul in TE.
  3. Provide a bacterial plate if submitting a BAC for injection.
  4. Provide a restriction map and sequence of the construct including size, vector, promoter, gene of interest and/or reporter gene.
  5. Provide gel documentation of the purity and concentration of the DNA construct.
  6. Obtain IACUC approval for your project before submission of the DNA.
  7. Obtain space within the Animal Resources Center for housing founder animals.
  8. Submit a “service request form”.

 Fees for Transgenic Mouse Production service

  • $3000 will be charged upon identifying 2 founder mice.
  • $3500 will be charged for injection of a BAC construct.
  • $2500 when using YAC DNA. IMPORTANT. Currently we are unable to prep YAC DNA for investigators. We can still inject this type of DNA if provided by the investigator using an approved purification protocol. In addition, we will only guarentee the injection of 200 embryos, not 2 founders as stated with plasmid DNA. Finally, it will be the investigators responsiblity to genotype the mice.
  • An additional $1000 will be charged if investigator requests C57Bl/6 embryos for injection.
